5-HT1A receptor

Also called: serotonin 1A receptor

A serotonin receptor that 5-MeO-DMT binds more strongly than any other, which researchers suspect helps explain why it feels different from DMT. DMT and psilocin bind it too, more weakly.

The 5-HT1A receptor is one of the serotonin receptors. It matters most for 5-MeO-DMT: a 2022 review of two binding studies on cloned human receptors reports that 5-MeO-DMT binds 5-HT1A most strongly, with 300 to 1,000-fold selectivity over 5-HT2A (Reckweg et al., 2022).

That tilt may shape the experience. In rodents, several of 5-MeO-DMT’s effects are reduced by a 5-HT1A blocker, and the review notes that 5-HT1A activation tends to lower heart rate and blood pressure while 5-HT2A activation tends to raise them (Reckweg et al., 2022). An observational study suggests its sparse visual imagery “may relate to its unusually high agonism” at 5-HT1A (Timmermann et al., 2025), which is a hypothesis.

DMT binds 5-HT1A as well (Barker, 2018). In a 2026 study of 12 volunteers, pretreatment with pindolol, a 5-HT1A blocker, intensified the effects of a sub-hallucinogenic DMT infusion, and blood pressure rose with the combination (Zahid et al., 2026). It is one small study and needs repeating.
